    <p>Blood contains cell free DNA (cfDNA), and cell free mitochondrial DNA(cf-mtDNA) is able to be easily detected by real time PCR. The present study investigated the relationship between the amount of cf-mtDNA in plasma and physiological conditions of cows. In addition, relationship between preg-nancy following artificial insemination and concentration of cf-mtDNA in the corresponding plasma. Significantly positive relationship was found between concentration of LDH and cf-mtDNA, and negative relationship was found between concentration of triglyceride and cf-mtDNA. There was no significant difference in the amount of cf-mtDNA between pregnant or non-pregnant cows following artificial insemination.</p>

    <p> Embryo transfer uses embryos developed <i>in vivo</i> or <i>in vitro</i> for cattle production, however there is a difference in the quality of the embryos obtained by the two methods. This study addresses the differences in gene expression between blastocysts developed <i>in vitro</i> and <i>in vivo</i>. <i>In vivo</i> blastocysts were flushed from the uteri of super-ovulated cows and blastocysts developed <i>in vitro</i> were derived from <i>in vitro</i> matured and fertilized embryos. The same batch of frozen bull sperm was used for insemination and <i>in vitro</i> fertilization. Blastocysts were then subjected to RNA sequencing. Differentially expressed genes upregulated in <i>in vitro</i> blastocysts were annotated to focal adhesion, extracellular matrix (ECM)-receptor interaction, and PI3K-Akt signaling and the genes that were upregulated in <i>in vivo</i> blastocysts were annotated to oxidation-reduction processes, mitochondrion organization, and mitochondrial translation. Although the total cell number of the two types of blastocysts was similar, the mitochondrial quantity (determined by mitochondrial DNA copy numbers and expression levels of TOMM20), and ATP content in the blastocysts were lower in <i>in vivo</i> blastocysts compared with those developed <i>in vitro</i>. In conclusion, RNAseq revealed differential molecular backgrounds between <i>in vitro</i> and <i>in vivo</i> developed blastocysts and mitochondrial number and function are responsible for these differences.</p>

    <p>Enzootic bovine leukosis is the most common neoplastic disease in cattle caused by the bovine leukemia virus (BLV). Previous studies have shown that allele type in the <i>DRB3</i> region of the bovine major histocompatibility complex (bovine leukocyte antigen: <i>BoLA</i> in cattle) is associated with resistance or susceptibility to infectious diseases. The Japanese brown (JBR) cattle have a genetic background of small population size, and thus, may exhibit different susceptibility to diseases compared with other cattle breeds. In the present study, we analyzed the diversity of the <i>BoLA-DRB3</i> gene and attempted to clarify the genetic composition of the Kumamoto and Kochi strains belonging to the JBR.</p><p>Allele typing of the <i>BoLA-DRB3</i> gene of 184 JBR detected 16 and 13 alleles in the Kumamoto and Kochi strains, respectively. The analysis of the observed (<i>H</i>o) and expected (<i>H</i>e) heterozygosity of the JBR based on allele frequencies, as well as <i>F</i>is (using <i>H</i>o and <i>H</i>e), which indicates the inbreeding coefficient within a population, showed each population to have different <i>BoLA-DRB3</i> gene diversities. Consequently, the degree of genetic difference among the populations (<i>F</i>st) was analyzed based on the allele frequencies of the <i>BoLA-DRB3</i> gene. The results showed the JBR populations to be genetically closer to the native Philippine and Myanmar cattle populations than the Japanese black, Japanese Shorthorn, Holstein or Jersey population. Furthermore, the proportions of cattle with at least on allele, which was reported to be associated with low proviral load (PVL) on infection of BLV, were 15.7% (20/127) and 63.1% (37/57) in Kumamoto and Kochi strain, respectively, and showed a significant difference. In the future, the direct evaluation of the <i>BoLA-DRB3</i> gene allele types and PVL in BLV-infected individual cattle is warranted. In addition, the PVL of BLV-infected JBR of the Kumamoto strain is known to be lower than that of other breeds in Japan. Therefore, it is necessary to investigate the possibility of a different mechanism on PVL regulation from allele types.</p><p>The results of <i>BoLA-DRB3</i> gene diversity and allele frequency analysis indicate that the JBR has a different genetic background from the Japanese black and the Shorthorn populations and possesses low PVL-associated alleles. Further studies are needed to analyze the relationship between PVL and <i>BoLA-DRB3</i> gene in JBR to clarify the susceptibility of JBR to BLV.</p>

  • Predicting of molecules mediating an interaction between bovine embryos and uterine epithelial cells Reviewed

    <p> Embryo-maternal reproductive tract interactions are pivotal for successful pregnancy. The present study predicted the molecules modulating embryo-uterine communication by comparing two sets of differentially expressed genes (DEGs): DEGs in uterine epithelial cells (UECs) collected from the uterus with and without blastocysts and DEGs between blastocysts developed <i>in vivo</i> and <i>in vitro</i>. Cows were subjected to super ovulation (SOV), followed by insemination or non-insemination at estrus (SOV + AI and SOV cows). Seven days after estrus, the uterus was flushed to collect UECs, and the presence of blastocysts in the uterus was confirmed. UECs were subjected to RNA-Sequencing (RNA-Seq) to identify DEGs. Publicly available RNA-Seq data of <i>in vivo</i> and <i>in vitro</i> developed bovine blastocysts were used to determine DEGs. Then, using ingenuity pathway analysis, activated- and inhibited-upstream regulators (USRs) for UECs in blastocysts were compared with those for blastocysts developed <i>in vivo</i>. RNA-Seq of UECs revealed that the DEGs were associated with immune response and cell adhesion pathways. The activated and inhibited USRs of UECs derived from SOV+ AI cows overlapped with the activated and inhibited USRs of blastocysts developed <i>in vivo</i>. Overlapping activated USRs include leukemia inhibitory factor, interleukin 6, fibroblast growth factor-2, transforming growth factor beta-1, and epidermal growth factor. In conclusion, the present study predicted the molecules that potentially mediate communication between the developing embryo and the uterus <i>in vivo</i> and prepare the uterus for pregnancy.</p>

    <p>Bovine leukemia virus (BLV) is an important pathogen associated with enzootic bovine leukosis. In this study, we performed PCR and sequencing analysis to characterize BLVgp51 sequences from formalin-fixed paraffin-embedded (FFPE) specimens made from 1974 to 2000 and successfully obtained BLV proviral genome sequences from 94% of the analyzed samples. Furthermore, from these samples, we reconstructed eight full-length and nearly full-length BLVgp51 sequences. These sequences were classified as BLV genotype 1, implying that genotype1 has already been circulating in Japan since the 1970s. In our results, the proviral DNA was detected in the 1970s, 1980s, and 1990s in the same manner, indicating that the detection of BLV proviral genome depends on storage conditions rather than storage period. The sequences obtained in this study provide direct insights into BLV sequences before 2000, which serves as a good calibrator for inferring ancient BLV diversity.</p>

    繁殖雌ウシ飼養農家34戸、延べ231頭を対象に、発情発見方法についてアンケート調査を実施した結果、外陰部の発赤・粘液・乗駕を発情徴候として観察している農家が多数を占め、受胎牛の示した発情徴候は、外陰部の発赤・透明粘液が多数であり、不受胎牛では、挙動不審・透明粘液・乗駕の割合が多い傾向にあった。高受胎率農家では透明粘液を徴候としていたが、低受胎率農家では、行動量増加を徴候としていた。

    過酸化物還元能力を有する乳酸菌を分離し、母豚の周産期および授乳期の子豚に投与する事で安全性と機能性に富むプロバイオティクスの確立を検討した。その結果、乳酸菌は投与期間中、子豚の増体量を促進する働きがあると考えられた。また、母豚への投与は子豚の肥育終了までその効果が持続することが示唆された。さらに、肥育成績では、子豚への乳酸菌投与群が非投与群に対し、枝肉重量、歩留および等級において良好な成績であった。以上のことから、哺乳期間中の乳酸菌投与は、肥育成績に良好な影響を及ぼすことが示唆された。

    ウシではβ-カロテンの利用はよく知られているが、キサントフィル類は吸収しにくいと言われている。そこで、ウシがアスタキサンチンを吸収できるか検討した。血液からカロテノイドを抽出し、高速液体クロマトグラフィーで分析した結果、アスタキサンチン給与後の供試牛の血液において高速液体クロマトグラフィーのチャート上アスタキサンチンの位置にピークを認めた。さらにアスタキサンチンの存在を明確にするため、カロテノイド抽出液をシリカゲルクロマトグラフィーにかけてキサントフィル画分を分取、濃縮して高速液体クロマトグラフィーにより分析し、アスタキサンチンのピークを確認した。以上の結果より、ウシはアスタキサンチンを吸収できると考えられた

    養豚飼料に利用する茶屑のペレット化において、作製条件の違いが豚の嗜好性に及ぼす影響を検討した。嗜好性改善に利用する糖蜜の添加量は3%と50%、また糖蜜3%にトウモロコシ粉砕物10%を添加した3種ペレットの硬度、嗜好性を比較したところ、糖蜜の含量が高いほど硬度は高くなり、トウモロコシの添加は低くなった。ペレットのみでの嗜好性テストではトウモロコシの添加が好まれたが、飼料と同時に給与した場合は、糖蜜だけの添加でも良好な採食性が認められた。

    冬季に保育する肉用牛子牛に脂肪酸カルシウムまたは脂肪酸カルシウムとカルニチンを補給した際の発育を無添加の対照牛の発育と比較した。脂肪酸カルシウムあるいは脂肪酸カルシウムとカルニチンを補給することで対照区に比べ発育が良好となる傾向が示され、なかでもカルニチンを補給した方が良好となる傾向にあり、脂肪酸カルシウムで補給されたエネルギーの利用がカルニチンで促進されたと推察された。
