Several types of lectin domains that specifically recognize chitin have been discovered in plants. One such domain, the hevein domain, also known as CBM18, contains eight cysteine and glycine residues at conserved positions in 40 amino acid residues. It works alone, arranged in tandem, or in combination with other domains. Tomato lectin is a chimeric lectin composed of four hevein domains and extensin-like domains similar to the plant cell wall glycoprotein extensin. It has been used for tissue staining and the fractionation of sugar chains owing to its specificity against poly-N-acetyllactosamine. In this minireview, the author summarizes the current literature on the chitin-binding lectins of plants and discuss the role of tomato lectin.

[Objective] Lectins are carbohydrate-binding proteins that agglutinate red blood cells. Our group has identified two lectin genes (TL-F and TL-L) encoding chitin-binding lectins that share similar molecular features but exhibit different expression profiles. We investigated the distribution and genetic relationships of both genes among inbred tomato lines. [Results] Of the 7 inbred tomato lines, 5 lines were homozygotes of TL-F (F/F) and 2 lines were homozygotes of TL-L (L/L). No heterozygotes (F/L) were found. The genotypes of F2 generations derived from the hybrid F1 (F/L) lines were separated following the law of segregation. These results suggest that TL-F and TL-L genes are alleles from the same locus.

【Objective】Plant glandular trichomes (GT) are specialized epidermal tissues with subcuticular storage space that accumulate volatile organic compounds (VOCs). The small lipid transfer protein (LTP) is thought to be involved in the transport of VOCs from the secretory cells across the cell wall to the storage space of GTs. The GTs of hop (Humulus lupulus L.) cones, termed lupulin glands, accumulate in a high content of essential oil that imparts flavor to beer. To understand the molecular basis for terpene accumulation in lupulin glands, we analyzed the role of LTP expressed in hop lupulin. 【Results】 (1) Transcriptome analysis by RNAseq showed several non-specific LTP1 genes were expressed in hop lupulin, and their deduced sequences were obtained. (2) Three LTP1 proteins were purified from the extract of lupulin. Among them, the content of HlLTP1.1 was the highest. Histochemical analysis showed that HlLTP1.1 localizes in the cell walls of secretory cells of lupulin glands. (3) The gene expression level of HlLTP 1.1 was correlated with the essential oil content among several hop cultivars. These results suggest that LTP plays a role in transporting terpenes to the storage space of the lupulin gland.
