鉄は葉緑体の電子伝達系の構成因子であり、安定した光合成を可能にする。仮に鉄が長期的に欠乏すると、葉緑体の電子伝達が異常となり、最終的に光酸化ストレスによって細胞死が起こる。本研究では、鉄欠乏オオムギの光合成機能の最適化を担う必須因子HvLhcb1.12に着目した。HvLhcb1.12は集光性アンテナタンパク質の一種であるが、そのN末端側には多数のリン酸化部位が存在する。本遺伝子をイネに導入した結果、過剰光条件下での収量が野生型のイネより約25%増加し、光酸化ストレスの回避に貢献することが示された。

オオムギ地上部に存在する新規鉄欠乏耐性機構の解明

植物において鉄は光合成、呼吸、およびその他の酸化還元反応や代謝に必須な役割を担っている。本研究では鉄欠乏に適応したオオムギの性質に着目し、1.オオムギ体内での鉄移行システム、2.オオムギ細胞内鉄貯蔵システム、3.鉄欠乏下での光合成の維持能力という3つの観点から解析を進めた。この中で、マイクロアレイ解析を実施し、鉄欠乏耐性に関与しうる遺伝子を絞り込んだところ、クロロフィルa/b結合蛋白質をコードする遺伝子Lhcb1のホモログ遺伝子が大幅に発現上昇していることが明らかになった。そこで、鉄欠乏誘導性のLhcb1遺伝子を単離し、さらに、Lhcb1の蓄積に異常を示すオオムギ変異株を用いて解析を進めた。この結果、Lhcb1は特に鉄欠乏で発生する光ストレスの回避に必須な因子であることが明らかになった。本研究により植物の鉄栄養研究と光合成研究にまたがって新規な鉄欠乏適応機構の一端が明らかにされた。
